    Highly variable Vitis microsatellite loci for the identification of Pinot Noir clones

    Nineteen new microsatellite loci of Vitis were elaborated by following the procedure of tagging an SSR-enriched library. Primers for these VRG markers were used for genotyping grapevines. Only the markers VRG 1, VRG 2, VRG 4, VRG 7, VRG 9, VRG 10, VRG 15 and VRG 16 show heterozygous alleles and Mendelian segregation. Other VRG loci such as VRG 5, VRG 6, VRG 11, VRG 12 VRG 13 and VRG 17 produce a multiallelic profile and some of them show distorted segregation. Variability of the VRG loci is rather high as compared to other grapevine SSR markers. Stable VRG markers such as VRG 16 can be useful for the identification of cultivars. Highly variable VRG microsatellites could be successfully applied to trace polymorphism within the variety Pinot Noir. Clones of Pinot Noir could be differentiated using these markers. By applying the PhyQuest program, a dendrogramm showing the genetic divergence within Pinot Noir clones was constructed.This project was funded by the Ministry of Agriculture and Forestry in Vienna.     DON’T SLIP ON THE ICO – A TAXONOMY FOR A BLOCKCHAIN-ENABLED FORM OF CROWDFUNDING

    Blockchain is rapidly evolving and there is an increasing interest in the technology in both practice and academia. Recently, a blockchain use case called Initial Coin Offering (ICO) draws a lot of attention. ICO is a novel form of crowdfunding that utilizes blockchain tokens to allow for truly peer-to-peer investments. Although, more than 4.5 billion USD have been invested via ICOs, the phenomenon is poorly understood. Scientific research lacks a structured classification of ICOs to provide further insights into their characteristics. We bridge this gap by developing a taxonomy based on real-world ICO cases, related literature, and expert interviews. Further, we derive and discuss prevailing ICO archetypes. Our findings contribute to theory development in the field of ICOs by enriching the descriptive knowledge, identifying design options, deriving ICO archetypes, and laying the foundation for further research. Additionally, our research provides several benefits for practitioners. Our proposed taxonomy illustrates that there is no one-size-fits-all model of ICOs and might support the decision-making process of start-ups, investors and regulators. The proposed ICO archetypes indicate how common ICOs are designed and thus might serves as best practices. Finally, our analysis indicates that ICOs represent a valid alter-native to traditional crowdfunding approaches

    Tarzan and chain: exploring the ICO jungle and evaluating design archetypes

    The phenomenon of a blockchain use case called initial coin offering (ICO) is drawing increasing attention as a novel funding mechanism. ICO is a crowdfunding type that utilizes blockchain tokens to allow for truly peer-to-peer investments. Although more than \$7bn has been raised globally via ICOs as at 2018, the concept and its implications are not yet entirely understood. The research lags behind in providing in-depth analyses of ICO designs and their long-term success. We address this research gap by developing an ICO taxonomy, applying a cluster analysis to identify prevailing ICO archetypes, and providing an outlook on the token value market performance for individual archetypes. We identify five ICO design archetypes and display their secondary market development from both a short-term and a long-term perspective. We contribute to an in-depth understanding of ICOs and their implications. Further, we offer practitioners tangible design and success indications for future ICOs

    Antiinflammatory Therapy with Canakinumab for Atherosclerotic Disease

    Background: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. Methods: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. The primary efficacy end point was nonfatal myocardial infarction, nonfatal stroke, or cardiovascular death. RESULTS: At 48 months, the median reduction from baseline in the high-sensitivity C-reactive protein level was 26 percentage points greater in the group that received the 50-mg dose of canakinumab, 37 percentage points greater in the 150-mg group, and 41 percentage points greater in the 300-mg group than in the placebo group. Canakinumab did not reduce lipid levels from baseline. At a median follow-up of 3.7 years, the incidence rate for the primary end point was 4.50 events per 100 person-years in the placebo group, 4.11 events per 100 person-years in the 50-mg group, 3.86 events per 100 person-years in the 150-mg group, and 3.90 events per 100 person-years in the 300-mg group. The hazard ratios as compared with placebo were as follows: in the 50-mg group, 0.93 (95% confidence interval [CI], 0.80 to 1.07; P = 0.30); in the 150-mg group, 0.85 (95% CI, 0.74 to 0.98; P = 0.021); and in the 300-mg group, 0.86 (95% CI, 0.75 to 0.99; P = 0.031). The 150-mg dose, but not the other doses, met the prespecified multiplicity-adjusted threshold for statistical significance for the primary end point and the secondary end point that additionally included hospitalization for unstable angina that led to urgent revascularization (hazard ratio vs. placebo, 0.83; 95% CI, 0.73 to 0.95; P = 0.005). Canakinumab was associated with a higher incidence of fatal infection than was placebo. There was no significant difference in all-cause mortality (hazard ratio for all canakinumab doses vs. placebo, 0.94; 95% CI, 0.83 to 1.06; P = 0.31). Conclusions: Antiinflammatory therapy targeting the interleukin-1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid-level lowering.     Breeding of fungal resistant varieties derived from Grüner Veltliner by chromosomal selection

    Traditional cultivar Grüner Veltliner is the most appreciated vine in Austrian viticulture. Due to organic growing the demand for mildew resistance within the same wine profile has increased. Cross breeding can provide such new genotypes which combine traits from different sources by parenthood. Linkage of traits with chromosomes or markers allows to predict some aspects of the phenotype. Equipped with chromosomal assisted selection the development of new varieties could be much easier and faster. On the base of two segregating populations derived from crosses of Grüner Veltliner with Malverina and Seyval blanc we could define correlation of chromosomes with some traits. Mainly ampelographic descriptors and resistance against mildew could be aligned. As a quality parameter of the wine Rotundone analyses were performed and could be attributed to chromosome 5 and 9. Selection supported by the composition of the parental chromosomes enables breeding with some arguments of design. The limits for free choice were the availability of sufficient different genotypes with a broad spectrum of chromosomal combinations. Recently released descendent cultivar Donauveltliner was selected due to the high rate of Traminer alleles

    Quantitative trait loci analysis of erinose (Eriophyes vitis Pgst.) on a genetic map Welschriesling x Sirius

    Three biological races of Erinose (Eriophyes vitis Pgst. sin Colomerus vitis Pgst.) commonly attack vineyards every year. A genetic map Welschriesling x Sirius (Mandl et al. 2006) was used as the basis of QTL (quantitative trait loci) analysis for Erinose. The symptoms were evaluated in five consecutive years in 92 individuals from this segregating population and other OIV traits related to the density of hairs in different tissues were also scored. QTL analysis was performed on all data using Plabqtl version 1.1. A major QTL for Erinose sensitivity was identified and reproducible every year on linkage group 9d close to the SSR marker VMC5c1. Two additional smaller QTLs could also be identified in linkage groups 5 and 17. The fact that some OIV traits related to the density of hairs were linked to the same group of the genetic map, is also discussed.Peer Reviewe
